Single-cell response to KCl stimulation
In the single-cell response to KCl stimulation, the isolated cell model is configured and parameterised with an applied concentration clamp protocol.
You can change the parameters of stimulation in the component clamp_para. You can also bypass the parameters in the component Para and define your own parameters.
The simulation experiment can be obtained by loading the corresponding SED-ML document into OpenCOR and executing the simulation. We use the Python script simFig4 to perform the simulation for the following figures.
We set the intracellular K + concentration Kin = 158.4mM and the initial value of the open state probability of K + channels wi = 0.5 during the simulation.
Dynamics when Kout = 4.7mM
Figure [1] shows the oscillating dynamics of Ca2 + concentrations and the cell membrane potential, when the extracellular K + concentration Kout = 4.7mM.

Single-cell response to KCl stimulation
We set the extracellular K + concentration Kout = 4.7mM before t = 10s and Kout steps up to 80 mM at t = 10s. Since the reversal potential for K + current is associated with Kout, the reversal potential vK in original equation (13) is also changed at t = 10s.
Figure [2] shows the simulation result where there is an initial surge of ci when we use the initial values from the cited model (Parthimos et al. 1999).

We tried different initial values and Figure [3] shows a few traces compared to the original figure. Although the closest orange trace has some overlap with the original data, there is still discrepancy between them. We think this might have something to do with the intracellular K + concentration value Kin = 158.4mM or the holding extracellular K + concentration Kout = 4.7mM before t = 10s.
